EasyGeno Single Assembly Cloning kit (VI202-02)
The EasyGeno Single-Fragment
Recombinational Cloning Kit enables rapid directional cloning of a single DNA
fragment into any linearized vector, based on sequence homology between the fragment
and vector ends. Through meticulous design and rigorous testing, the included 2×
EasyGeno Single Assembly Mix contains enzymatic activity that mediates recombination
and linkage between the homologous regions of the vector and the insert starting
from the 3′ ends of the double-stranded DNA. This results in a seamless
closed-circular plasmid that can be directly transformed into competent cells for
clone selection.
This method overcomes the limitations of restriction enzyme sites: only the terminal
sequences of the linearized vector need to be introduced at the 5′ ends of the PCR
primers used to amplify the insert. This generates 15–25 bp overlaps between the
insert ends and the corresponding vector ends, allowing directional recombination of
the fragment into the vector. The recombination reaction is performed rapidly under
isothermal conditions (50 ℃ for 15 minutes). Combined with flexible PCR product
handling options and a rapid transformation protocol, the entire process—from DNA
sample preparation to transformation and plating—can be completed in just a few
hours.
EasyGeno Assembly Cloning Kit (VI201)
The EasyGeno Assembly Cloning Kit enables
rapid directional cloning of single or multiple DNA fragments into any linearized
vector, based on sequence homology between the fragments and the vector. The
thoughtfully formulated one-tube master mix—2× EasyGeno Assembly
Mix—facilitates recombination and ligation between the linearized
vector and insert fragments at homologous regions, producing seamless
closed-circular plasmids. These can be directly transformed into competent cells for
clone selection.
This method eliminates dependence on restriction enzyme sites: directional
recombination of the target fragment into the vector is achieved simply by
incorporating 15 bp homologous ends of the linearized vector onto both ends of the
insert via PCR. The recombination reaction is performed rapidly under isothermal
conditions (50 ℃ for 15 minutes), with a positive clone rate exceeding
95%.
In addition to single-fragment cloning, the kit also supports simultaneous
directional assembly of multiple fragments into a vector, enabling high-throughput
construct generation
